Markers for immuno-electron microscopy

چکیده
The development of techniques for electron microscopic studies of biological specimens has made it possible to visualize fine structural details of intact cells in ultrathin sections. The high magnification power of electron microscopes, along with refined techniques for biological specimen preparation, has enabled a wealth of informations with respect to normal and pathological fine structure of organs. Yet, what is usually detected is a map of distribution of electrons scattered by the macromolecular contents of the ultrathin section of the cells. Because spatial and temporal aspects of cellular processes and the functional or evolutionary significance of the complexity of higher organisms cannot be elucidated simply by comparing fine structures, specific detection methods at the molecular level are required for the identification and localization of individual components, quite analogous to light microscopy for which specific stains have been developped which absorb or emit characteristic optical radiation. For the latter, immunohistological stains with fluorochrome or enzyme labelled antibodies have proved most specific and are most generally used. In electron microscopy, such specific stains will have to scatter electrons in characteristic manner.

Gold particle quantification in immuno-gold labelled sections for transmission electron microscopy
We briefly outline an algorithm for accurate quantification of specific binding of gold particles to fixed biological tissue samples prepared for immuno-transmission electron microscopy (TEM). The algorithm is based on existing protocols for rational accounting of colloidal gold particles used in secondary antibodies for immuno-gold labeling.
متن کاملNano-fEM: protein localization using photo-activated localization microscopy and electron microscopy.
Mapping the distribution of proteins is essential for understanding the function of proteins in a cell. Fluorescence microscopy is extensively used for protein localization, but subcellular context is often absent in fluorescence images. Immuno-electron microscopy, on the other hand, can localize proteins, but the technique is limited by a lack of compatible antibodies, poor preservation of mor...
متن کاملDetecting and Segmenting Nanodiscs in Immuno-Electron Micrographs
Nanodiscs are soluble nanoscale phospholipid bilayers with applications in drug delivery and the study of membrane proteins, for example. They can be imaged using electron microscopy, along with immunogold markers indicating locations of proteins of interest. We describe and evaluate methods for automatically detecting and segmenting nanodiscs in electron micrographs. The detection method modif...
متن کاملInvestigation of the ultrastructure of yeast using cryo-immuno-electron microscopy
Yeast has been a crucial model system for the investigation of a multitude of cellular processes and many approaches have been used to study these, including genetics, biochemistry and fluorescence microscopy approaches. The morphological analyses of these organisms by electron microscopy (EM), however, have remained limited because of the lack of techniques leading to satisfactory ultrastructu...
متن کاملDemonstration of correlative atomic force and transmission electron microscopy using actin cytoskeleton
In this study, we present a new technique called correlative atomic force and transmission electron microscopy (correlative AFM/TEM) in which a targeted region of a sample can be observed under AFM and TEM. The ultimate goal of developing this new technique is to provide a technical platform to expand the fields of AFM application to complex biological systems such as cell extracts. Recent adva...
متن کاملImmuno-analysis of microparticles: probing at the limits of detection
Microparticle (MP) research is clouded by debate regarding the accuracy and validity of flow cytometry (FCM) as an analytical methodology, as it is influenced by many variables including the pre-analytical conditions, instruments physical capabilities and detection parameters. This study utilises a simplistic in vitro system for generating MP, and through comparative analysis with immuno-electr...
